A major milestone in the field of male contraception has been reached with the success of YCT 529, an experimental male contraceptive pill that passed its first human safety trial. Unlike traditional methods, this groundbreaking pill temporarily stops sperm production without affecting hormones, offering a promising and balanced alternative for couples.

How Does YCT-529 Work?

Developed by YourChoice Therapeutics, YCT 529 works differently from hormonal birth control pills. It inhibits a protein called Retinoic Acid Receptor Alpha (RAR-α), which plays a vital role in sperm production. The key advantage? It does not alter testosterone levels.

According to early studies, men can fully regain fertility once they stop taking the pill—making it a reversible and hormone-free contraceptive option.

99% Effective in Animal Trials

  • In Mice: Just four weeks of YCT-529 use drastically reduced sperm production and successfully prevented 99% of pregnancies during mating.
  • In Primates: A noticeable decline in sperm count was observed within two weeks of usage.
  • Fertility Recovery: Male mice regained fertility within 6 weeks; primates within 10–15 weeks after stopping the pill.
  • No side effects were reported in these preclinical trials.

Results from First Human Safety Trial (Phase 1a)

The initial trial involved 16 healthy men aged between 32 and 59 who had previously undergone vasectomy—a surgical procedure that blocks sperm transport. This ensured the drug wouldn’t unintentionally affect fertility.

Participants received different doses of YCT 529 (10, 30, 90, or 180 mg) or a placebo. Some took the drug while fasting; others took it after a high-fat, high-calorie meal. The pill was quickly absorbed and reduced in concentration by half within 2–3 days—ideal for once-daily dosing.

Importantly, no adverse effects were noted on:

  • Heart rate
  • Hormone levels
  • Inflammation
  • Sexual desire
  • Mental health

Why YCT-529 Is Better Than Hormonal Options

Traditional hormonal male contraceptives often cause mood swings, reduced libido, weight gain, and acne. Since YCT-529 doesn’t influence hormone levels, it offers a safer, cleaner, and more attractive solution.

What’s Next?

Encouraged by the positive outcome of Phase 1a, a Phase 1b/2a trial is now underway involving 50 men who will take YCT-529 for 28 or 90 days. This phase will evaluate its long-term safety and its impact on sperm count.

Reports indicate nearly half of all pregnancies worldwide are unplanned, and men currently have only two options—condoms or vasectomy. A pill like YCT 529 provides a shared responsibility model and significantly reduces the contraceptive burden on women.
